Anefficientandrapidprotocolhasbeendevelopedforextractingginkgolidesandbilobalide(terpene
trilactones)fromGinkgobilobaleaves.Theproceduretakesadvantageoftheextraordinarystabilityof
theterpenetrilactonestructuretoavarietyofchemicaltreatments,especiallyoxidation,despitethe
presenceofmultipleoxygenfunctions.Theprotocolinvolvesboilingtheaqueoutractofleaveswith
dilutehydrogenperoxide,extractionwithethylacetate,washingwithbasicsolutions,andcharcoal
filtrationtoyieldanoff-whitepowder,terpenetrilactonecontent6070%.Itislikelythatthehydrogen
peroxidetreatmentdegradestheundesiredleafconstituentsthatleadtointenseemulsificationduring
extractions.Furtherreversed-phasechromatographyoftheextractswithpolymericresinsremovesthe
undesirableginkgolicacidstoamountslessthan10ppm.Theextractsaresuitedforpureterpenetrilactone
preparation,enrichmentofterpenetrilactonecontentinnutraceuticals,andpreparationsoflow-flavonoid/
high-terpenetrilactonecontrolsinmedicinalstudies.Thefourginkgolides(ginkgolidesA,B,C,J)and
bilobalideisolatedfromtheextractwereidenticalinallrespectswithauthenticspecimens.
