生物化学实验指导——试用版 PAGE 21 PAGE PAGE 21 血清成分分析（家鸡、乌鸡血清） 作者：王志豪 李东 导师：吴明江 （温州大学生命与环境科学学院 浙江温州 325027） 摘 要 ：本次血清成分分析总共分三个试验进行操作。血清胆固醇的定量测定（邻苯二甲醛法），血清清蛋白与γ-球蛋白的分离与鉴定，血清谷丙转氨酶活性的鉴定及活力测定。通过试验了解并掌握邻苯二甲醛法测定血清总胆固醇，盐析法分离提纯蛋白质，并对蛋白质进行透析与浓缩，采用凝胶层析法分离重铬酸钾和蓝葡聚糖混合液，利用醋酸纤维素薄膜电泳法分离与鉴定血清清蛋白与γ-球蛋白，凝胶过滤层析及透析法脱盐与蛋白质浓缩的原理和方法。并以肝脏为原料，进行了谷丙转氨酶活力的鉴定。利用对鸡的血清分析来了解血清成分的区别，我们主要采用乌鸡与家鸡对照来进行试验。 关键词： 血清、肝脏、成分分析、蛋白质、谷丙转氨酶 Serum component analysis ( chicken, chicken serum ) Author: Wang Zhihao Lidong Tutor: Wu Mingjiang (College of Life and Environment Sciences, Wenzhou University, Wenzhou, Zhejiang 325027 ) Abstract: The analysis of serum components in a total of three test operation.
