三七总皂苷对泡沫细胞内ABCA1和LXRα mRNA表达的影响 李卓莹,何雪峰,张海港,贾 乙,李晓辉(第三军医大学药学院新药研究中心,重庆400038) 摘 要: 目的 观察三七总皂苷(panax notoginseng saponins, PNS)对泡沫细胞ABCA1和LXRα mRNA表达及脂质沉积的影响。方法 提取SD大鼠腹腔巨噬细胞纯化后,与高、中、低剂量PNS(80、40、20 μg/ml)及氧化低密度脂蛋白共孵育48 h。油红O染色观察细胞内脂质堆积情况,酶法测定细胞内总胆固醇(TC)、游离胆固醇(FC)、胆固醇酯(CE)的含量并计算CE/TC值。采用RT-PCR检测泡沫细胞ABCA1和LXRα mRNA表达情况。结果 模型组细胞中TC、FC、CE及CE/TC值均显著高于对照组(P0.05);与模型组相比,PNS高、中剂量组细胞内TC、FC、CE及CE/TC均显著降低(P0.05)。PNS高、中剂量组中ABCA1及LXRα mRNA的表达均显著高于模型组(P0.01)。结论 PNS可以上调ABCA1和LXRα mRNA表达并减少巨噬细胞源性泡沫细胞中脂质沉积。 三七总皂苷;泡沫细胞;ABCA1;LXRα Panax notoginseng saponins on mRNA expressions of ABCA1 and LXRα in foam cells LI Zhuo-ying, HE Xue-feng, ZHANG Hai-gang, JIA Yi, LI Xiao-hui (Research Center of New Drugs, College of Pharmacy, Third Military Medical University, Chongqing400038, China) Abstract: Objective To observe the effects of Panax notoginseng saponins (PNS) on mRNA expressions of ABCA1 and LXRα and lipid deposition in foam cells. Methods Peritoneal macrophages obtained from SD rats after purification were incubated with ox-LDL for 48 h to establish foam cell model. PNS (20, 40, 80 μg/ml, respectively) were given for intervention. The cellular lipid accumulation was examined by oil red staining. The cellular contents of total cholesterol (TC), free cholesterol (FC) and cholesterol ester (CE) and the ratio of CE/TC were detected by enzymic methods. The mRNA levels of ABCA1 and LXRα were determined by RT-PCR. Results The contents of TC, FC, CE and CE/TC ratio in model group were significantly increased compared with control group (P0.05). Compared with model group, 40, 80 μg/ml PNS reduced the contents of TC, FC, CE and CE/TC ratio remarkably (P0.05) and increased ABCA1 and LXRα mRNA expressions significantly (P0.01). Conclusion PNS reduces lipid deposition and upregulates ABCA1 and LXRα mRNA expressions in foam cells.
