苦参碱对溃疡性结肠炎大鼠肠粘膜细胞因子和自由基的影响

摘要 目的 探究苦参碱对溃疡性结肠炎大鼠肠粘膜细胞因子和自由基的影响机制。方法 TNBS)复制大鼠溃疡性结肠炎治疗结束后,取结肠,检测溃疡性结肠粘膜细胞中IL-1β、TNF-α、IL-4、IL-10等一系列细胞因子的水平;检测结肠粘膜细胞中SOD、MDA水平。做组织显微镜检查结果 1)与模型组相比,苦参碱和柳氮磺胺吡啶均能极显著降低IL-1β水平(p0.01);显著降低TNF-α水平(p0.01);极显著降低IL-4水平(p0.01);极显著降低IL-10水平(p0.01);2)与模型组相比,苦参碱和柳氮磺胺吡啶均能极显著升高结肠粘膜细胞SOD水平(P0.01);极显著降低MDA水平(P0.01);3)苦参碱和柳氮磺胺吡啶均能促进溃疡面愈合,减少病灶部位炎细胞浸润,水肿及纤维化。结论 苦参碱能明显抵抗溃疡性结肠炎炎性反应,增强机体免疫,通过调节肠粘膜细胞因子失衡和抑制粘膜细胞氧自由基的产生和抗氧化功能干预了UC发病过程。
关键词 苦参碱 溃疡性结肠炎 细胞因子 自由基Abstract Objective This study is to investigate the effect of matrine on the mechanism of cytokines and free radical in UC rats intestinal mucosa. Methods We made the model of ulcerative colitis by TNBS, and used the matrine to treat animals. After the treatment, animals were killed and colons were taken out, a series of cytokines IL-1β, TNF-α, IL -4, IL-10 were detected; The SOD and the MDA were detected too. Other colons were tanken to be done the microscopy, and tissue injury scores were caculated. Result 1)Compared with model group, matrine and sulfasalazine could significantly reduce the IL-1β levels (p0.01); significantly decrease TNF-α levels (p0.01); significantly reduce IL-4 levels (p0.01); significantly reduce IL-10 levels (p 0.01); 2) Compared with model group, matrine and sulfasalazine could significantly increase the level of SOD in colon mucosa cells (p 0.01); could significantly decrease MDA levels (p 0.01); 3) matrine and sulfasalazine could heal ulcer cells, reduce the lesion area of inflammatory cell infiltration, edema and fibrosis. Conclusion matrine could resist ulcerative colitis inflammatory response, enhance body immune, by regulating cytokine imbalance in the intestinal mucosal cells and inhibiting the free radical formation and antioxidant intervention in the pathogenesis of UC.
